From January 24th to 25th, 2024, the 8th China International Cosmetics Technology Forum of InnoCosme 2024 was successfully held in Shanghai. Hou Zengmiao, director of R&D of Zhanyan Bio, was invited to attend and gave a sharing titled "Exploration and Breakthrough of Zhanyan Recombinant Functional Protein Raw Materials", introducing the basic situation of Zhanyan Bio in the field of synthetic biology and its innovative progress in basic research.

The theme of the forum was "Biotechnology leads product innovation, insight into the market and opens a new consumption era", which brought together the frontier trends of the industry and invited experts and scholars to discuss and share the development progress of new raw materials and technological innovation breakthroughs.

Based on synthetic biology and consolidating the hard power of scientific research

In the fierce industry competition, the position of scientific research strength is increasingly prominent, and it has become an important factor in enterprise competition. Zhanyan Bio is based on synthetic biology, based on consumer insight and new consumption trends, tackling genetic engineering technology, exploring solutions to skin problems and protecting skin health.

Hou Zengmiao, director of research and development of Zhanyan biological raw materials, introduced at the forum that Zhanyan biological has rich experience in gene design, strain construction, fermentation and purification in the application of recombinant protein expression system of Pichia pastoris and Escherichia coli, and has established a production platform for recombinant functional protein strain construction (Pichia pastoris and Escherichia coli), small-scale production research, pilot-scale amplification research and large-scale fermentation and purification.

In addition, relying on two major genetic engineering research and development platforms, Denoheisi and InPuBoJi, Zhanyan Bio has deployed a large number of terminal applications in the fields of bioactive molecular synthesis biology technology, immobilized enzyme catalysis technology, high-density fermentation purification technology, key technology of recombinant protein anti-inflammatory and healing-promoting, biomedical material construction technology and patented raw material formulation technology, covering the whole industrial chain of bioactive raw materials, biomedical materials, medical terminal products and dermatological grade skin care products.

At present, Zhanyan Bio owns 113 patents, including 43 invention patents, 52 new utility patents and 18 appearance patents. Thanks to the related genetic engineering technology, Zhanyan Bio has realized the research, development and mass production of bioactive materials such as recombinant mussel mucin, recombinant humanized collagen, recombinant fibronectin, recombinant elastin and recombinant superoxide dismutase, and has been applied to many series of products.

Patented preparation technology adds recombinant mussel mucin to realize mass production

Mytilus mucin is very rare. Mytilus mucin acts on the damaged epidermis through film formation, thus playing the role of isolation and protection and achieving good repair effect. However, the traditional natural mussel mucin has low efficiency and the source of raw materials is at risk of being polluted. Easy to use but scarce, its name of "Ocean Gold" was born.

Hou Zengmiao, director of research and development of Zhanyan biological raw materials, said that Zhanyan biological research and development team used synthetic biology technology to break away from convention, established a controllable modification process of mussel mucin dopa in vitro with self-developed and self-produced high-activity tyrosinase, broke through the bottleneck of large-scale mass production of mussel mucin, and innovated endotoxin removal methods to control endotoxin in mass production process with minimum loss. Through various innovative technologies, the recombinant mussel mucin was successfully prepared, in which the purity was more than 95%, the dopa content was as high as 5.81%, far exceeding the requirements of the industry standard of 0.3%, and the endotoxin content was less than <0.5 EU/mg.

Based on its own biotechnological advantages and the functional application of recombinant mussel mucin, Zhanyan Bio has continuously studied recombinant mussel mucin and promoted its clinical application. Up to now, more than 20 papers have been published. The "recombinant mussel mucin for dressing" drafted by Denoheis, the research and development platform of Zhanyan Bio-genetic engineering, was officially implemented in 2023, and five patents related to recombinant mussel mucin have been authorized. Zhanyan Bio has become a pioneer in the research and development and production of recombinant mussel mucin.

A breakthrough was made in hydroxylation-recombinant humanized collagen III and recombinant elastin.

Compared with collagen extracted from animal sources, humanized collagen is derived from human collagen gene, which has better affinity and biocompatibility, higher purity and stability, and at the same time, it puts an end to virus pollution and allergic risk that may accompany animal source extraction. Zhanyan recombinant humanized collagen was prepared by advanced synthetic biology technology, and its core sequence peptide was 100% homologous to human collagen sequence, and it was rich in integrin binding sites such as KGD and RGD. After fermentation by Pichia pastoris, the purity of the final product was ≥95%, and the endotoxin content was < <0.05 EU/mg, which could meet the quality requirements of implantable medical raw materials. Up to now, Zhanyan Bio has achieved the mass production of recombinant humanized collagen III, I and IV, and the research and development of recombinant humanized collagen XVII has been completed, and the patents related to the preparation of recombinant humanized collagen have been granted.

Hydroxyproline is the basic feature of natural collagen, but there is no recombinant collagen containing hydroxyproline in the domestic market at present. After two years of technical research, Zhanyan Bio has broken the technical barrier that non-mammalian expression systems (e.coli and yeast) cannot be hydroxylated without P4H. Through the transformation of Pichia pastoris chassis cells and fermentation technology, the proline hydroxylation of recombinant humanized collagen is realized. At present, the hydroxyproline content in laboratory level can reach 12.279%, which is close to the level of natural human collagen hydroxyproline of 13%. The breakthrough of this technology has promoted the research level of domestic recombinant collagen to achieve a qualitative improvement, and also injected more powerful impetus into the development of domestic skin care products.

At the same time, Zhanyan Bio has also made a breakthrough in the research of recombinant elastin. The R&D team selected the functional core peptide of natural human elastin, took Pichia pastoris as the host bacteria, and obtained the mass-produced recombinant elastin through ultrafiltration, chromatography, endotoxin removal and other processes. The purity of recombinant elastin produced by this technology was ≥95%, endotoxin was < 0.05EU/mg, and HCP and HCD all met the requirements of medical materials standards. Recombinant elastin has remarkable activity of promoting cell proliferation and migration, especially in the anti-photoaging cell model, showing excellent anti-photoaging activity. Recombinant elastin can significantly reduce the activity of metalloproteinase MMP-1 and increase the expression of type I collagen and type III collagen, among which the expression of physiological elastin can be increased by 300%.

Based on biotechnology, Zhanyan Bio has continuously increased its investment in research and development, and made major breakthroughs in many fields such as functional protein raw materials.
